Will I be charged a penalty fee or interests for owing more money on an amended return ?

Yes, probably.  If you do the IRS will send a bill.

What makes no sense to me is that it said I would of avoided a penalty fee or interests if I had amended my return prior to April 15th. Turbotax did not make this option available. Even if I send a cheque for the new amount I owe due to my amendment, they will start charge a fee?

I assumed you were referring  to a prior years amended return.   Am amended 2020 return would have no penalty or interest if the tax was paid before 5/17.
