If I have never claimed American opportunity tax credit before during college, can I still claim it while I am in graduate school?

No, the AOTC is only available for the first four years of schooling.  You can still claim the Lifetime Learning Credit for graduate school.

There's one possible exception. Grad school in the fall does count if the student finished his undergrad degree in the spring, The technicality is that you had not completed the first four years of post-secondary education as of the beginning of the taxable year.
