Can i stop and resume later when I'm using the TurboTax Premier disc downloaded to my computer: that is, not TurboTax Online? File > Save, then close? Some other way?

Yes, you can save your return that way, or even when you simply close your return, you'll be asked if you wish to save it. See the screenshot below.
