My employer did not add $500 to a Roth 401k plan as was deducted from my paychecks and instead returned the money deducted to me via check. This change was noticed after my W-2 was issued and I filed taxes.

Do I need to file an amendment with the corrected W-2 that shows I did not contribute to a Roth 401k for $500?

No, if the only change on the corrected W-2 is in box 12, then nothing should change if you were to file an amended tax return.  Keep the corrected W-2 in your tax records.
