Do I have to sign Form 8453-PE as ERO to EFILE my Partnership return?
I have signed form 8453 as General Partner, do I also have to sign again as the ERO before I save the form to my return and efile it?

No, you do not have to sign it again as the ERO (and actually should not do so) since TurboTax (Intuit) is effectively the ERO.
