My wife's last name is hyphenated, TurboTax is saying "Please enter your spouse's last name, using only letters"

You will have to leave out the hyphen in order to complete and e-file the return.

 

While that is grammatically (and socially) incorrect, it is necessary. When you e-file your return, the information has to be specifically formatted so that the IRS and state processing systems can properly handle your return when it is received. One of the conditions of this formatting is that the data cannot contain special characters such as hyphens or dashes.
